|Abstract:||A multi-target Real-Time Polymerase Chain Reaction (RTi-PCR) analytical system as a detection tool for the unequivocal identification of EU approved and some unapproved Genetically Modified Plants has been developed in response to the constantly growing testing needs for GMO throughout the world. The methodology and format allow immediate implementation into laboratories’ routine work since the system uses an RTi-PCR 96-well platform, commonly used worldwide and adopted in most EU control laboratories. The use of ready-to-use plates, with a guaranteed low variability, facilitates comparability of results. The approach presented is considered flexible and may represent the starting point for a whole new set of applications. The system enables the event-specific simultaneous detection of 39 single-insert GMOs (and of derived stacked events). The performance of this novel system has been successfully confirmed by experimental testing conducted within the European Union Reference Laboratory for Genetically Modified Food and Feed (EURL-GMFF) and in collaboration with the European control laboratories (Querci et al, 2009). It was shown that as low as 0.045% of the respective GMO (expressed as haploid genome copies in a single species) can be detected. As such, the performance of this system is in full compliance with the method performance criteria set by the 'European Network of GMO Laboratories' (ENGL). New GMO specific targets are being included - as new methods become available – and a crop-specific formulation specifically for soybean and maize events has been already developed. The system contains event-specific methods to detect 6 GM soybean and 14 GM maize events and covers the full range of EU soybean and maize authorized GM events (date 15 July 2010). Here we discuss the rationale and comparative advantage of the selected strategy as well as the formulation, potentiality and flexibility of the system towards a harmonized approach in GMO analysis. The crop-specific RTi-PCR system for GM soybean and maize is used as an experimental example. Also, an indication is presented as to the development of future simplex (or combinatory) RTi-PCR platforms for GMO analysis with respect to production quality measures, minimal performance requirements and decision support systems.|
