Inter-individual variations: a challenge for the standardisation of complement activation assays
The role of the human immune system in pathologic responses to chemicals including nanomaterials was identified as a critical gap in current hazard assessment schemes. In the health sector, nanotechnology-based health products using liposome and lipid nanoparticles carriers are promising tools as drug delivery systems and received recently attention as vectors for SARS-COV 2 vaccines. However, the complexity of the human immune system, interspecies variations and even fluctuations in inter-individual responses makes the development and standardization of predictive in vitro tests challenging.
We investigated the variability of complement activation in sera of healthy donors and patients suffering from cancer, immunodeficiency and allergies upon exposure to small molecules and liposomes. We could demonstrate significant differences of the complement activation in pooled human serum compared to specific sera from healthy donors or donors with preconditions.
